Description
Proliferating cell nuclear antigen (PCNA), formerly known as cyclin, is a 36kD intranuclear acidic protein closely associated with proliferating cells. PCNA was originally identified in patients with systemic lupus erythematosus. It represents a component of DNA polymerase-b. The presence of PCNA is closely associated with the cell cycle. PCNA is expressed in high concentration in the late G1-phase before the DNA synthesis, and it reaches a maximum concentration in the S-phase. The level of PCNA declines during the G2 and M phases.

Applications:
Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested.

Recommended Dilutions:
Immunohistochemistry: 1:50-1:100 (Paraffin). Requires membrane permeabilization. Does not require protein digestion pre-treatment or antigen retrieval of paraffin sections.
Immunohistochemistry: Frozen sections
Flow Cytometry: 1:100 Use 10ul to label 10e6 cells in 100ul. Requires membrane permeabilization.
Western Blot: 1:1000
Optimal working dilutions to be determined by researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
