Description
PRSS8 is a member of the trypsin family of serine proteases. It can be found in seminal fluid and is highly expressed in prostate epithelia. The proprotein is cleaved to produce a light chain and a heavy chain which are associated by a disulfide bond. The carboxyterminal region contains a GPI anchor sequence, which anchors prostasin to the cell surface. Phospholipase activity is thought to liberate prostasin from the cell, and cleavage of the linker region also releases prostasin. PRSS8 has been proposed as a marker for ovarian cancer, where it is up-regulated, a marker for androgen-refractory prostate cancer, where it is down-regulated, and as an inhibitor of invasiveness in breast cancer.

Applications:
Suitable for use in ELISA and Western Blot. Other applications not tested.

Recommended Dilutions:
ELISA (Capture): 0.2-0.8ug/ml
Western Blot: 0.1ug/ml with the appropriate secondary reagents to detect mouse Prostasin. The detection limit for recombinant mouse Prostasin is ~20ng/lane under non-reducing and reducing conditions.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
