Specificity
Recognizes human Protein tyrosine kinase-like 7 at ca.86kD in colon tissue lysates. Species Crossreactivity: mouse, rat.
Description
Protein tyrosine kinase-like 7 (PTK7) is a member of the receptor tyrosine kinase family, otherwise known as Colon Carcinoma Kinase 4 (CCK4). PTK7 is highly expressed in the kidney, liver, lung, pancreas and melanocytes with weaker expression being detected in the thyroid gland, ovaries, brain, heart and skeletal muscle.

PTK7 may play a role in embryonic development. It is thought to function as a cell adhesion molecule and as a novel regulator of the planar cell polarity (PCP) pathway in vertebrates.

Studies also suggest that PTK7 may be involved in the tumor progression. A loss of PTK7 expression has been observed in melanoma cell lines, whereas increased levels of PTK7 have been detected in high-throughput acute myeloid leukemia samples. In addition, PTK7 is not expressed in normal colon but has been detected in colon carcinomas and as such is a potential marker for tumor progression.

Applications:
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.

Recommended Dilutions:
Immunohistochemistry (Paraffin): Requires antigen retrieval using heat treatment prior to staining paraffin sections. Sodium citrate buffer pH 6.0 is recommended.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.

Note: Applications are based on unconjugated antibody.
