Description
Stat3 is a key signaling molecule for many cytokines and growth-factor receptors (1). It is required for murine fetal development (2). Stat3 is constitutively activated in a number of human tumors (3, 4). It possesses oncogenic potential (5) and anti-apoptotic activities (3). Stat3 is activated by tyrosine phosphorylation at Tyr705, which induces dimerization, nuclear translocation and DNA binding (6, 7). Transcriptional activation seems to be regulated by serine phosphorylation at Ser727, apparently via the MAPK or mTOR pathways (8, 9). Stat3 isoform expression appears to reflect biological function: the relative expression levels of Stat3alpha (86kD) and Stat3beta (79kD) depend on cell type, ligand exposure or maturation stage of the cells (10). It is notable that Stat3beta lacks the serine phosphorylation site within the C-terminal transcriptional activation domain (8).

Applications:
Suitable for use in Western Blot, Immunoprecipitation and Flow Cytometry. Other applications not tested.

Recommended Dilution:
Western Blot: 1:1000
Immunoprecipitation: 1:100
Flow Cytometry: 1:200
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
