Description
MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250, 000.

TRF2 is a ubiquitously expressed protein that is implicated in the control of telomere length. TRF2, like TRF1 contains a Myb-related DNA binding motif. It binds to duplex TTAGGG repeats and is localized to all human telomeres in metaphase chromosomes. TRF2 is thought to protect chromosome ends by maintaining the correct structure at telomere termini. The use of mutant forms of TRF2 has implicated a role for TRF2 in the prevention of senescence in primary human cells. Recently, it has been shown that inhibition of TRF2 resulted in apoptosis in a subset of mammalian cell types.

Applications:
Suitable for use in Western Blot and has been reported to demonstrate cell nuclear staining in Immunocytochemistry. Other applications have not been tested.

Recommended Dilution:
Western Blot: 0.5-2ug/ml detects TRF2 from a RIPA lysate of human Jurkat cells. TRF2 may be detected as a doublet in some cell lysates. Jurkat cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with T2400 (1ug/ml). Proteins were visualized using an IgG (HRP) goat anti-mouse *and a chemiluminescence detection system.
Optimal dilutions to be determined by the researcher.

Positive Antigen Control:
J9000: Jurkat cell lysate. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.

Storage and Stability:
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.

Note: Applications are based on unconjugated antibody.
