TNF-α lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20˚ C. Reconstitute in sterile water to a concentration of 0.1 - 1.0 mg/mL. The antibody is stable for at least six weeks at 2-4˚ C when reconstituted.

Background
Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide stimulated macrophages, and causes tumor necrosis when injected into tumour bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia.

Application Note
Neutralization:
To yield one-half maximal inhibition [ND50] of the biological activity of hTNF-α (0.5 ng/mL), a concentration of < 0.1 μ g/mL of this antibody is required.
ELISA:
In a sandwich ELISA (assuming 100 μ L/well), a concentration of 2.0-4.0 μ g/mL of this antibody will detect at least 1000 pg/mL of recombinant human TNF-α when used with our biotinylated antigen affinity purified anti-human TNF-α (XP-5282Bt) as the detection antibody at a concentration of approximately 0.5-1.0 μ g/mL.
Western blot:
To detect hTNF-α by Western Blot analysis this antibody can be used at a concentration of 0.5-1.0 μ g/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant hTNF-α is 0.5-1.0 ng/lane, under reducing conditions and 0.25-0.50 ng/lane, under non-reducing conditions.

The monoclonal antibody was produced in BALB/c X ICR F1 mice using recombinant hTNF-alpha as the immunizing antigen.
