The human Interleukin 6 (IL-6) kit is a sandwich ELISA. The capture antibody is a monoclonal human IL-6 antibody pre-coated onto the 96-well strip plates provided in the kit. Human test samples and standards of known IL-6 concentration are added to these wells and allowed to complex with the bound IL-6 antibody. A biotinylated human IL-6 polyclonal antibody is then added. This detection antibody binds to the antigen thus completing the sandwich. After washing, an enzyme Avidin-Biotin-Peroxidase complex (ABC) is added which binds to the second antibody. The peroxidase substrate TMB is added to induce a coloured reaction product. The intensity of this coloured product is directly proportional to the concentration of IL-6 present in the samples. The purpose of this kit is the in-vitro quantitative determination of human IL-6 in samples such as sera, plasma, tissue lysates and cell culture supernates. This kit has been configured for research use only and is not to be used in diagnostic or clinical procedures.

IL-6 is a cytokine with many biological functions. Some of its roles include the final differentiation of B-cells into Ig-secreting cells, induction of meloma and plasmacytoma growth, induction of nerve cell differentiation, and in hepatocytes, induction of acute phase reactants. IL-6 is secreted and is N- and O-glycosylated post translationally. Genetic variations in IL-6 can be correlated with bone mineral density and low bone mineral density is a risk factor for osteoporosis. Defects in IL-6 are associated with juvenile rheumatoid arthritis and at least one polymorphism in IL-6 makes HIV-infected men susceptible to Kaposi sarcoma which is a relatively rare malignant tumour.
