Background
Smad2 (also known as Mothers against decapentaplegic homolog 2, Mothers against DPP homolog 2, Mad2, hMAD-2 or hSMAD2) is a member of the Smad family of proteins which are similar to the gene products of the Drosophila gene 'mothers against decapentaplegic' (Mad) and the C. elegans gene Sma. SMAD proteins are signal transducers and transcriptional modulators that mediate multiple signaling pathways. This protein mediates the signal of the transforming growth factor (TGF)-beta, and thus regulates multiple cellular processes, such as cell proliferation, apoptosis, and differentiation. This protein is recruited to the TGF-beta receptors through its interaction with the SMAD anchor for receptor activation (SARA) protein. In response to TGF-beta signal, this protein is phosphorylated by the TGF-beta receptors. The phosphorylation induces the dissociation of this protein with SARA and the association with the family member SMAD4. The association with SMAD4 is important for the translocation of this protein into the nucleus, where it binds to target promoters and forms a transcription repressor complex with other cofactors. This protein can also be phosphorylated by activin type 1 receptor kinase, and mediates the signal from the activin. Alternatively, spliced transcript variants have been observed for this gene. SMAD2 may act as a tumor suppressor in colorectal carcinoma. It positively regulates PDPK1 kinase activity by stimulating its dissociation from the 14-3-3 protein YWHAQ which acts as a negative regulator. SMAD2 may be associated with diseases such as Keloids and Ureteral Disease. This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal region of human Smad2 protein.

Purification
This affinity purified antibody is directed against human Smad2 protein. The product was affinity purified from monospecific antiserum by immunoaffinity chromatography. A BLAST analysis was used to suggest cross-reactivity with Smad2 protein from human, mouse and rat based on 100% homology with the immunizing sequence. Reactivity against homologues from other sources is not known.  Also, the antibody is Smad2 specific, and reactivity to other Smad proteins (specifically Smad1, Smad3, Smad4, and Smad7) is not detected in over-expressed cell lysates (Personal Communication, Kathleen Flanders, CCR-NCI, Bethesda, MD).
