Storage Conditions
Storage at -20°C is recommended, as aliquots may be taken without freeze/thawing due to presence of 50% glycerol. Stable for at least 1 year at -20°C. After date of receipt, stable for at least 1 year at -20°C.
Observed Molecular Weight
97295 MW
Clonality
Polyclonal
Application notes
Specific for endogenous levels of the ~95 kDa dynamin protein phosphorylated at Ser774. Labels the purified protein phosphorylated in vitro by cdk5 but not by PKC. Does not cross react with other purified substrates of cdk5 (e.g. amphiphysin and synapsin). Immunolabeling is blocked by ?-phosphatase treatment.
Application Details
WB: 1:1000
ICC: 1:1000
Manufacturer - Gene Name
DNM1
Gene Full Name
Dynamin-1
Background
Dynamin is a member of a group of nerve terminal proteins called dephosphins that regulate synaptic vesicle endocytosis (Cousin et al., 2001; Graham et al., 2002; Tsuboi et al., 2002). Cyclin dependent protein kinase 5 phosphorylates dynamin at Ser-774 and Ser-778 that are the phosphorylation sites on dynamin phosphorylated in vivo (Tan et al., 2003). Phosphorylation of Ser-774 by GSK3 has recently been shown to control activity-dependent bulk endocytosis of synaptic vesicles (Clayton et al., 2010).
Immunogen
Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser774 of rat dynamin, conjugated to keyhole limpet hemocyanin (KLH). Immunogen species is Rat.
Contents
10 mM HEPES (pH 7.5), 150 mM NaCl, 100 µg per ml BSA and 50% glycerol.
Purification
Prepared from pooled sheep serum by affinity purification via sequential chromatography on phospho and non-phosphopeptide affinity columns.
Concentration
0.5-1mg/ml, actual concentration vary by lot. Use suggested dilution ratio to decide dilution procedure.
Phospho Site
Phosphorylated, Ser774

Protein Name
Dynamin-1
Protein Function
Microtubule-associated force-producing protein involved in producing microtubule bundles and able to bind and hydrolyze GTP. Most probably involved in vesicular trafficking processes. Involved in receptor-mediated endocytosis (By similarity).
Subcellular Localization
Cytoplasm. Cytoplasm, cytoskeleton. Microtubule-associated (PubMed:9725914). Isoform-specific localization (PubMed:9725914).
