Human tPA Active ELISA Kit

Tissue Plasminogen Activator (PLAT/tPA) is a secreted serine protease which converts the proenzyme Plasminogen to Plasmin, a fibrinolytic enzyme. tPA is synthesized as a single chain which is cleaved by Plasmin to a two chain disulfide linked protein. This enzyme plays a role in cell migration and tissue remodeling. Increased enzymatic activity causes hyperfibrinolysis, which manifests as excessive bleeding, decreased activity leads to hypofibrinolysis which can result in thrombosis or embolism. Functionally active tPA will form a covalent complex with the biotinylated Human PAI1 which is bound to the avidin on the plate. After appropriate washing steps, polyclonal Anti-Human tPA primary antibody binds to the captured tPA. Excess antibody is washed away and bound polyclonal antibody is then reacted with the HRP-conjugated secondary antibody. Following an additional washing step, TMB is used for color development at 450 nm. The amount of color development is directly proportional to the concentration of active tPA in the sample.
