Storage Conditions
The lyophilized product, though stable at room temperature for up to 3 weeks, should be best stored desiccated at -80C for more than 1 year or at 2-8C for about 10 months. Reconstituted protein should be used immediately or stored in undiluted working aliquots at -20C. For long term storage it is recommended to add a carrier protein (0.1% BSA). Avoid repeated freeze/thaw cycles.

Description
CD14 acts as a receptor for endotoxin (LPS) and is expressed strongly on monocytes, macrophages, and weakly at the surface of neutrophils. CD14 is anchored to cells by linkage to glycosylphosphatidylinositol (GPI) and functions as a high affinity receptor of LPS-LBP-complexes. Furthermore, CD14 is present in a soluble form in human serum, urine and other bodily fluids, acts at physiological concentrations as an LPS agonist, and has at higher concentrations an antagonizing effect in cell activation. The myeloid differentiation antigen CD14 acts as the major receptor for bacterial LPS. The dominant form of the recombinant wild type CD14 is the 50 kDa protein. The human CD14 was produced from human CD14 transfected CHO-cells in serum free medium. Before transfection, the complete human CD14-cDNA was amplified by PCR and cloned into expression vector p-POL-DHFR.
Formulation
Lyophilized from a concentrated protein solution (0.25 mg/ml) in PBS, pH 7.2.
Reconstitution
Centrifuge vial prior to opening. Add 40 ul sterile distilled water to the vial. Further dilutions should be made with PBS or other buffers.
Biological Activity
Up to 20 ug/ml CD14 inhibits binding of FITC-LPS (0.5 ug/ml) to 6 x 105 CD14+ CHO cell transfectants as determined by flow cytometry.
