Description
The peptide RRRADDSDDDDD can be used as a substrate for CK2 (casein kinase 2) in, in vitro, kinase assays. It is phosphorylated by CK2 holoenzyme with a Km of 20 uM and a Vmax of 232.1 nmol/min/mg. Casein kinase 2 (CK2) is a ubiquitous pleiotropic proliferation-associated serine/threonine protein kinase. Similar to protein kinase A, CK2 is a tetramer made up from two different subunits alpha2beta2. The catalytic alpha-subunit corresponds to the C-subunit of PKA, the non-catalytic beta-subunit is unique and differs from the R-subunit of PKA in all known features. Recently it was shown that the beta-subunit is trifunctional: (i) it confers stability to the holoenzyme, (ii) it increases enzyme activity, and (iii) it determines substrate specificity.
