Background
beta-Secretase (BACE1) is an aspartyl protease involved in the cleavage of amyloid precursor protein (APP). BACE1 is a transmembrane protein with two active site aspartate residues in its extracellular protein domain. BACE1 may operate functionally as a dimer.

Generation of amyloid-beta peptide requires two sequential cleavages of amyloid precursor protein (APP). Extracellular cleavage of APP by BACE1 creates a soluble extracellular fragment and a cell membrane-bound fragment. Cleavage of a membrane bound fragment by gamma-secretase releases the intracellular domain of APP and yields amyloid-beta peptide.

Levels of BACE1enzymatic activity appear to be elevated in age related, late-onset Alzheimer's potentially suggesting a gene regulatory mechanism.
