Application Note:
ELISA: Use at an assay dependent dilution. IHC-P: Use at a concentration of 1 µ g/ml. IP: Use at a concentration of 5 µ g/ml. WB: Use at a concentration of 0. 1 µ g/ml. Detects a band of approximately 12 kDa representing parvalbumin from rat cerebellum. Parvalbumin protein is relatively small and therefore it is recommended that electrophoresis be performed using tricine-SDS-PAGE gels and transferred to a nylon membrane. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. Parvalbumin (PV) is a calcium binding protein expressed in specific muscle fibers and fast-firing neurons. PV consists of a single unbranched chain of linked amino acids and belongs to a larger group of EF hand proteins. Studies have demonstrated that parvalbumin acts in the decay of calcium in the contraction/ relaxation cycle of fast twitch muscles. This data has shown a positive correlation between the rate of relaxation and the concentration of parvalbumin. Parvalbumin is also expressed in a specific population of GABAergic interneurones which are thought to play a role in maintaining the balance between excitation and inhibition in the cortex as well as the hippocampus. In amyotrophic lateral sclerosis (ALS) patents parvalbumin immunoreactivity is specifically absent from neuron populations lost early in ALS.
Function:
In muscle the calcium-binding protein parvalbumin is thought to be involved in muscle relaxation. Mass Spectrometry: Mass=11927; Mass_error=0. 76; Method=Electrospray; Range=2-110; Source=PubMed:10036163; .
Miscellaneous:
This protein binds two calcium ions.
Similarity:
Belongs to the parvalbumin family.
Similarity:
Contains 2 EF-hand domains.
