Application Note:
IHC-P: Working dilution ready-to-use for 10 minutes at room temperature. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Citrate buffer pH 6. 0 for 10 minutes followed by cooling at RT for 20 minutes. 18-272-196480 shows nuclear cytoplasmic and cell membrane staining. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. Cellular
Localization:
Cytoplasmic and Nuclear Ubiquitin is a highly conserved protein of about 8. 5 kDa molecular weight which has a normal role in the targeting of proteins for proteolytic degradation. To perform this function the protein to be degraded is first covalently attached to the C terminus of ubiquitin and the ubiquitinated complex is then recognized by a complex of degradative enzymes. Interestingly ubiquitin also becomes covalently bonded to many types of pathological inclusions which appear to be resistant to normal degradation. Therefore ubiquitin antibodies are very useful for studies of these inclusions. For example the neurofibrillary tangles and paired helical filaments diagnostic of Alzheimer\' s disease Lewy bodies seen in Parkinson\' s disease and Pick bodies found in Pick\' s disease are all heavily ubiquitinated and can be readily visualized with ubiquitin antibodies.
Function:
Protein modifier which can be covalently attached to target lysines either as a monomer or as a lysine-linked polymer. Attachment to proteins as a Lys-48-linked polymer usually leads to their degradation by proteasome. Attachment to proteins as a monomer or as an alternatively linked polymer does not lead to proteasomal degradation and may be required for numerous functions including maintenance of chromatin structure regulation of gene expression stress response ribosome biogenesis and DNA repair.

Ubiquitin is synthesized as a polyubiquitin precursor with exact head to tail repeats the number of repeats differ between species and strains. In some species there is a final amino-acid after the last repeat here in human a Val. Some ubiquitin genes contain a single copy of ubiquitin fused to a ribosomal protein (either L40 or S27a).
