Localization:
Type I membrane protein Vascular Endothelial Growth Factor Receptor 2 (VEGFR 2 also known as KDR / FLK 1) is a 200 kDa member of a receptor tyrosine kinase family whose activation plays an essential role in a large number of biological processes such as embryonic development wound healing cell proliferation migration and differentiation. As with other growth factor receptors upon ligand binding VEGFR 2 dimerizes and is autophosphorylated on multiple tyrosine residues. These phosphotyrosine sites are involved in regulation of intrinsic tyrosine kinase activity or serve as binding sites for SH2 domains and phosphotyrosine binding (PTB) domains of downstream signaling proteins. Phosphorylation of Tyrosine 1214 is reported to play a critical role in regulation of receptor autophosphorylation and stimulation of intrinsic tyrosine kinase catalytic activity.
Function:
Receptor for VEGF or VEGFC. Has a tyrosine-protein kinase activity. The VEGF-kinase ligand/receptor signaling system plays a key role in vascular development and regulation of vascular permeability. In case of HIV-1 infection the interaction with extracellular viral Tat protein seems to enhance angiogenesis in Kaposi\' s sarcoma lesions.
