Immunogen:	Peptide sequence C-DEANQRATKMLGSG corresponding to the C-terminus of SNAP-25 (NP_003072. 2 NP_570824. 1).
Specificity Note:	This product detects an epitope within the C-terminal (CT) of human synaptosomal-associated protein 25kD (SNAP-25). SNAP-25 is a presynaptic plasma membrane protein which is essential for Ca2+-regulated exocytosis of neurotransmitters. SNAP-25 has been shown to bind to and negatively modulate Ca2+ channels increasing the speed of neurotransmitter release. SNAP-25 is expressed by neurons and neuroendocrine cells as two predominant isoforms; SNAP-25a and SNAP-25b. SNAP-25a is expressed early in development whereas SNAP-25b expression is linked to synaptogenesis.
Antiserum Preparation:	Antisera to human SNAP-25 were raised by repeated immunisations of goats with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

Function:	t-SNARE involved in the molecular regulation of neurotransmitter release. May play an important role in the synaptic function of specific neuronal systems. Associates with proteins involved in vesicle docking and membrane fusion.
Subunit:	Part of the SNARE core complex containing SNAP25 VAMP2 and STX1A. This complex binds CPLX1. Interacts with TRIM9 RIMS1 SNAP25BP and HGS. Binds STXBP6. Found in a ternary complex with STX1A and VAMP8. Found in a complex containing SYT1 SV2B and syntaxin-1 (By similarity).
Subcellular Location:	Membrane; Lipid-anchor. Cell junction synapse synaptosome. Cell membrane; Lipid-anchor. Note=Membrane association requires palmitoylation.
Tissue Specificity:	Neurons of the neocortex hippocampus piriform cortex anterior thalamic nuclei pontine nuclei and granule cells of the cerebellum.
Ptm:	Palmitoylated. Cys-85 appears to be the main site and palmitoylation is required for membrane association (By similarity).
Miscellaneous:	When cloned and expressed in Eschericia coli where protein palmitoylation does not occur Cys-85 Cys-88 Cys-90 and Cys-92 in the protein sequence readily form an iron-sulfur cluster instead.
