Genway ID:
GWB-59C7C9
Clone:
RPA34-20
Isotype:
IgG2a
Immunogen:
Replication protein A purified from U293 cells.
Specificity:
Replication protein A (Ab-3) will react with both the complexed (phosphorylated) form of the protein as well as the free (unphosphorylated) 34 kDa subunit. Replication protein A can be used as a marker for cell division.
Positive Control:
HeLa or U293 cells or colon carcinoma tissue
Target:
RPA34
Localization:
Nuclear
Concentration:
0. 1 mg/ml
Purification Note:
From hybridoma culture supernatant Storage
Buffer:
PBS 0. 1mg/ml BSA no preservatives added
Application Note:
For ICC: Use at an assay dependent dilution. For IF: Use at a concentration of 2-3 µ g/ml. For IHC-P: Use at a dilution of 2-3µ g/ml. For IP: Use at a dilution of 1µ g/reaction. For WB: Use at a dilution of 4-6µ g/ml. Not tested in other applications. Optimal dilutions/concentrations should be determined by the researcjer. This antibody can be used as a marker for proliferation. Reacts with both the complexed (phosphorylated) form of the protein as well as the free (unphosphorylated) subunit. For paraffin sections pretreat with heat or use pressure cooker method. Target protein
Molecular Weight:
34kD Cellular
Localization:
Nuclear RPA is involved in DNA replication repair and recombination. Human RPA is a stable heterotrimer of 70 kDa 32-34 kDa and 11-14 kDa subunits (RPA70 RPA32 and RPA14 respectively). RPA is required for the SV40 large tumor antigen-catalyzed unwinding for SV40 DNA and stimulates DNA polymerase alpha and delta.
Function:
Required for DNA recombination repair and replication. The activity of RP-A is mediated by single-stranded DNA binding and protein interactions.
Subunit:
Heterotrimer of 70 32 and 14 kDa chains. The DNA-binding activity may reside exclusively on the 70 kDa subunit. Binds to SERTAD3/RBT1. Interacts with TIPIN.
Subcellular Location:
Nucleus. Note=Also present in PML nuclear bodies. Redistributes to discrete nuclear foci upon DNA damage.
Ptm:
Phosphorylated in a cell-cycle-dependent manner (from the S phase until mitosis). Phosphorylated by ATR upon DNA damage which promotes its translocation to nuclear foci. Can be phosphorylated in vitro by PRKDC/DNA-PK in the presence of Ku and DNA and by CDC2.
