Isotype:
IgG1
Immunogen:
Full length protein (Human).
Antigen Species:
Human
Positive Control:
HeLa cells
Target:
ORC1
Localization:
Nuclear.
Concentration:
1 mg/ml Storage
Buffer:
Phosphate-buffered saline pH 7. 2 containing 1% BSA and 0. 09% sodium azide
Western Blotting:
use at a concentration of 1 / 1250 - 1 / 2500. Not tested in other applications. Optimal dilutions / concentrations should be determined by the end user. By Western blot this antibody detects a band of 120 kDa which corresponds to the predicted molecular weight of ORC 1. Cellular
Localization:
Nuclear Myeloma: Sp2/0-Ag14 DNA replication in Saccharomyces cerevisiae requires the action of a multisubunit complex of six proteins known as the origin recognition complex (ORC). The identification of higher eukaryotic homologs of several ORC components suggests a universal role for this complex in DNA replication. ORC1 a sub-unit of the Origin Recognition Complex (ORC) is involved in the initiation of DNA replication. In human cells expression of ORC1 has been shown to be low in quiescent cells but dramatically induced upon stimulation of cell growth. In contrast expression of the HsOrc2 gene does not appear to be similarly regulated. HsORC1 a recently identified human protein related to yeast Orc1p and Cdc6p / Cdc18p may be a component of the replication initiation complex in human cells.
Function:
Component of the origin recognition complex (ORC) that binds origins of replication. It has a role in both chromosomal replication and mating type transcriptional silencing. Binds to the ARS consensus sequence (ACS) of origins of replication in an ATP-dependent manner.
Subunit:
ORC is composed of six subunits. Interacts with CDC6 and MYST2/HBO1.
Subcellular Location:
Nucleus.
Similarity:
Belongs to the ORC1 family.
Similarity:
Contains 1 BAH domain.
