Application Note:
For IP (Native with protein A): Use at a dilution of 1:400 protein lysate. For WB: Use at a dilution of 1:200. For IHC-P: Use at a dilution of 1:200. Not tested in other applications. Optimal dilutions/concentrations should be determined by the researcher. X-Ray Repair Cross Complementing (XRCC 1) plays a role in excision repair of DNA after ionizing irradiation. XRCC 1 binds to DNA ligase III through the C-terminal 96 amino acids and to DNA polymerase beta through the N-terminal half. In testis XRCC 1 is expressed at high levels. Cells with a mutation of this gene exhibit decreased single strand break repair and reduced recombination repair. They show increased double strand breaks and sister chromatid exchange is increased up to 10-fold. The XRCC 1 might serve as a scaffold protein during base excision-repair.
Function:
Corrects defective DNA strand-break repair and sister chromatid exchange following treatment with ionizing radiation and alkylating agents.
Subunit:
Homodimer. Interacts with polynucleotide kinase (PNK) DNA polymerase-beta (POLB) and DNA ligase III (LIG3). Interacts with APTX and APLF.
Subcellular Location:
Nucleus. Note=Accumulates at sites of DNA damage.
Ptm:
Phosphorylation of Ser-371 causes dimer dissociation. Phosphorylation by CK2 promotes interaction with APTX and APLF.

Carriers of the polymorphic Gln-399 allele may be at greater risk for tobacco- and age-related DNA damage.
