Specificity:
Appears to recognise the human pre-synaptic protein SNAP-25; an antigen of 26-27kD molecular weight. It will recognise SNAP-25 fusion protein from COS cells but not with the fusion protein from bacterial systems. Research studies have used this antibody to study the distribution of synaptic changes in the hippocampus of patients with medically refractory temporal lobe epilepsy. (1 3 & 8).

Application Note:
For IHC-P: Use at a dilution of 1:2 000 - 1:5 000. This product does not require protein digestion pre-treatment of paraffin sections. This product does not require antigen retrieval using heat treatment prior to staining of paraffin sections. Not tested in other applications. Optimal dilutions/concentrations should be determined by the researcher. Myeloma: Spleen cells from immunised BALB/c mice were fused with cells of the NSO myeloma cell line.
Function:
t-SNARE involved in the molecular regulation of neurotransmitter release. May play an important role in the synaptic function of specific neuronal systems. Associates with proteins involved in vesicle docking and membrane fusion.
Subunit:
Part of the SNARE core complex containing SNAP25 VAMP2 and STX1A. This complex binds CPLX1. Interacts with TRIM9 RIMS1 SNAP25BP and HGS. Binds STXBP6. Found in a ternary complex with STX1A and VAMP8. Found in a complex containing SYT1 SV2B and syntaxin-1 (By similarity).

Membrane; Lipid-anchor. Cell junction synapse synaptosome. Cell membrane; Lipid-anchor. Note=Membrane association requires palmitoylation.
Tissue Specificity:
Neurons of the neocortex hippocampus piriform cortex anterior thalamic nuclei pontine nuclei and granule cells of the cerebellum.
Ptm:
Palmitoylated. Cys-85 appears to be the main site and palmitoylation is required for membrane association (By similarity).
Miscellaneous:
When cloned and expressed in Eschericia coli where protein palmitoylation does not occur Cys-85 Cys-88 Cys-90 and Cys-92 in the protein sequence readily form an iron-sulfur cluster instead.
