Genway ID:
GWB-A0AB1B
Subclass:
IgG2a
Clone:
a-SM1
Specificity:
SM1 reacts exclusively with ?-smooth muscle actin which is typical for vascular and visceral smooth muscle cells. but which is also present in myofibroblasts. The epitope that is recognized by ?-SM1 is Ac-EEED. ?-SM1 is useful for immunocytochemistry. immunohistochemistry on frozen and paraffinembedded tissues. immunoblotting. immunoelectron microscopy and ELISA. Optimal antibody dilution should be determined by titration; recommended range is 1:1000 ? 1:10. 000 for immunohistochemistry with avidin-biotinylated horseradish peroxidase complex (ABC) as detection reagent. and 1:10. 000 ? 1:50. 000 for immunoblotting applications. Product
Note:
Each vial contains 100 ?l 1 mg/ml purified antibody in PBS containing 0. 09% sodium azide. Among the six actin isoforms described in mammals. two are found in virtually all cells (?? and ??cytoplasmic). two are detected in smooth muscle cells (?? and ?-smooth muscle) and two are present in striated muscles. one predominantly in skeletal (?-skeletal) and one in cardiac (?-cardiac) muscle cells. These actin isoforms differ slightly in their Nterminus and the sequences of each of them are perfectly conserved in higher vertebrates. Alphasmooth muscle actin is abundant in vascular and visceral smooth muscle cells. In addition. it has also been shown to appear in stress fibers of fibroblastic cells during pathological situations involving contractile phenomena such as wound healing and fibrocontractive diseases
Function:
F-actin cross-linking protein which is thought to anchor actin to a variety of intracellular structures. This is a bundling protein (By similarity).
Subunit:
Homodimer; antiparallel. Interacts with MYOZ2 PDLIM2 TTID and LPP (By similarity).
Subcellular Location:
Cytoplasm cytoskeleton. Cytoplasm myofibril sarcomere Z-disk. Note=Colocalizes with MYOZ2 and PPP3CA at the Z-line of heart and skeletal muscle.
Similarity:
Belongs to the alpha-actinin family.
Similarity:
Contains 1 actin-binding domain.
Similarity:
Contains 2 CH (calponin-homology) domains.
Similarity:
Contains 2 EF-hand domains.
Similarity:
Contains 4 spectrin repeats. Skalli. O. Ropraz. P. . Trzeciak. A. . Benzonana. G. . Gillessen. D. . and Gabbiani. G. (1986).
