Intended Use: The Human Retinol Binding Protein (RBP) test kits are a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring RBP in serum or plasma of Human\' s. Principle of the test: The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the RBP present in sample reacts with the anti-RBP antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound sample proteins by washing anti-RBP antibodies conjugated with horseradish peroxidase (HRP) are added. These enzyme-labeled antibodies form complexes with the previously bound sample RBP. Following another washing step the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate 3 3' 5 5' -tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of RBP in the sample tested; thus the absorbance at 450 nm is a measure of the concentration of RBP. The quantity of RBP in the test sample can be interpolated from the standard curve constructed from the standards and corrected for sample dilution. Storage and Stability: The expiration date for the package is stated on the box label. 1. DILUENTThe 5X Diluent Concentrate is stable until the expiration date. The 1X working solution is stable for at least one week from the date of preparation. Both solutions should be stored at 4-8 degree C. 2. WASH SOLUTIONThe 20X Wash Solution Concentrate is stable until the expiration date. The 1X working solution is stable for at least one week from the date of preparation. Both solutions can be stored at room temperature (16-25C) or at 4-8 degree C. 3. ENZYME-ANTIBODY CONJUGATEUndiluted horseradish peroxidase anti-HX conjugate should be stored at 4-8 degree C and diluted immediately prior to use. The working conjugate solution is stable for one day. 4. CHROMOGEN-SUBSTRATE SOLUTIONThe Substrate Solution should be stored at 4-8 degree C and is stable until the expiration date. 5. STOP SOLUTIONThe Stop Solution should be stored at 4-8 degree C and is stable until the expiration date. 6. ANTI-MOUSE IgA ELISA MICRO PLATEAnti-Mouse IgA coated wells are stable when stored at 4-8 degree C in the sealed foil pouch with desiccant pack until the expiration date. Unseal Microtitier Pouch and remove all strips and wells that will be used in the assay. Keep remaining strips and wells inside the sealed pouch with desiccant. 7. MOUSE IgA STANDARDSThe lyophilized Mouse IgA Calibrator should be stored at 4 degree C or frozen until reconstituted. The reconstituted calibrator should be aliquoted out and stored frozen (multiple freeze-thaws should be avoided). The working standard solutions should be prepared immediately prior to use and are stable for up to 8 hours. 8. INDICATIONS OF INSTABILITYIf the test is performing correctly the results observed with the standard solutions should be within 20 % of the expected values. Limitation of the Test: 1. Reliable and reproducible results will be obtained when the assay procedure is carried out with a complete understanding of the information contained in the package insert instructions and with adherence to good laboratory practice. 2. Factors that might affect the performance of the assay include proper instrument function cleanliness of glassware quality of distilled or deionized water and accuracy of reagent and sample pipettings. Total IgA in bronchoalveolar lavage fluid (BALF) of MME-exposed (mixture of microbial extracts) mice. 2. Aspergillus fumigatus [American Type Culture Collection (ATCC) 14109] Penicillium marneffei (ATCC 201013) Cladosporium herbarum (ATCC 201090) Candida albicans (ATCC 14053) Staphylococcus epidermidis (clinical isolate) and Escherichia coli (clinical isolate) were used for preparation of the mixture of microbial extracts (MME). Experiment Steps: 1. Prepare the cell-free BALF samples from five mice from the MME-treated group and the control group before infection (day 38). 2. Quantify protein content of individual samples using a BCA protein assay kit. 3. Concentrate all samples to a protein concentration of 200 ug/ml by ultracentrifugation. 4. Quantify total IgA in BALF samples using a mouse IgA ELISA Quantitation kit ( Biotech San Diego CA) following the manufacturer' s instructions. 5. Repeat the same experiment three times.
