Background
Enolase (2-phospho-Dglycerate hydrolase) is a cytoplasmic enzyme that is responsible for converting 2-phosphoglycerate to phosphoenolpyruvate. Three members: Enolase 1, Enolase 2, and Enolase 3, which are also termed alpha, gamma, and beta enolase, respectively. They exist as several dimeric isoenzymes including alphaalpha, alphabeta, betabeta, alphagamma, and gammagamma. The alphaalpha and gammagamma isoenzymes are abundant in neurons and neuroendocrine cells, and therefore, they are also designated as neuron specific enolase (NSE). Human Enolase 2 is 434 amino acids (aa) in length. It shares 83% aa identity with human enolases 1 and 3 and 99% with its mouse orthologue. Studies have shown that a variety of neurologic disorders such as stroke, traumatic brain injury, multiple sclerosis, and Alzheimer’s disease initiated release of Enolase 2 from injured cells and elevated serum levels of Enolase 2.

Assay principle
This Rat NSE ELISA Kit is based on standard sandwich enzyme-linked immunosorbent assay technology. Rat NSE specific antibody has been precoated onto 96-well plate. The test samples and the HRP conjugated Rat NSE specific detection antibody are added to the wells subsequently and then followed by washing the plate. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic Stop Solution. The density of yellow is proportional to the Rat NSE amount of sample captured in plate.
