Lectenz Bio, originally also known as Glycosensors and Diagnostics, is a biotechnology company specializing in glyco-research and bioprocessing. Lectenz Bio grew out of a research group and has specialized teams in the scientific fields of high performance computing and wet lab science, in addition to experts from industry. Their interdisciplinary collaboration has enabled the company to find patented solutions for glyco-research. Their user-friendly Lectenz® and GlycoSense™ technology platforms are available for this purpose.

What is glycan research all about?

Glycans are defined as a variety of monosaccharides linked by a glycosidic bond. A well-known example is cellulose, in which the D-glucose molecules are linked via a ß-1,4-glycosidic bond. Glyco-research is therefore concerned with the study and modification of glycans. The samples can consist of a wide variety of materials such as carbohydrates, glycoproteins, glycolipids, serum, tissue, lysates, cell culture supernatant, antibodies, or even glycobiology standards. Thus, glyco in a broader sense describes the carbohydrate portion of a given molecule. A number of techniques are used for the study including (enzymatic) glycan removal, glycan detection, and structural glycobiology and glycan profiling.

The difficulty in analysis and synthesis of glycoderivatives is that their structure can be very complex and heterogeneous.

This research aims to gain a better understanding of disease and its progression through the analysis of the glycosylated molecules involved, such as proteins. The knowledge gained can be used for the development of diagnostics and drugs, as well as for the development of vaccines.

Products

The Lectenz® product line represents a new class of lectin-like, enzyme-derived glycan target affinity reagents. The highly pure recombinant proteins were developed by computer-assisted directed evolution. It offers advantages over natural lectins in that it can bind glycan structures extremely specifically and offers improved properties in rapid detection and enrichment of glycoconjugates.

Application areas

Glycan detection

The SiaFindTM Lectenz® kits are used for the specific detection, separation or enrichment of sialoglycans. These are commonly found in glycoconjugates (glycoproteins, glycolipids and oligo- or polysacchrides). The products are based on a sialic acid affinity reagent.

The following products are available:

  • SiaFind™ Pan-Specific Lectenz® Kit (sialoglycans ending in Sia α-2,3-gal, Sia α-2,6-gal, and Sia α-2,8-sia). It has high affinity and specificity for sialoglycans independent of binding (pan-specific).
  • SiaFind™ Alpha 2,3-Specific Lectenz® Kit (sialoglycans ending in Neu5Ac-α-2,3-Gal) It has high affinity and specificity for α-2,3-linked sialic acids on glycans. However, it does not bind effectively to branched sialylated epitopes such as sialyl Lewis A/X.
  • SiaFind™ Alpha 2,6-Specific Reagent Kit (sialoglycans ending in Sia-α-2,6-Gal) It is suitable as a primary probe for ELISA, WB and immunohistochemistry.

Glycan removal

Our partner Lectenz Bio offers a range of glycan-cleaving enzymes. For example, α-2,3 sialidase, cleaves terminal α-2,3-linked sialic acid. PNGase F-II cleaves N-linked glycans with or without nuclear fructose, both α-1,6 and α-1,3 linkages.

Glycan profiling

The GlycoSense™ Array Kit is used for glycan profiling. The array consists of lectins conjugated to multiplex beads that bind terminal monosaccharides to glycoproteins. This can be used to determine the glycosylation pattern of glycoproteins using a flow cytometer. The array does not provide a complete glycosylation profile, but changes in several important glycan features can be detected.

The kit provides an alternative for analyses where mass spectrometry is not necessary or practical. The near real-time results obtained bring the advantage of direct monitoring of glycosylation, during protein expression and glycoenzyme reactions.
