Like cathepsin B, H, and L, cathepsin S belongs to the group of lysosomal cysteine proteinases. Cathepsin S is found primarily in the spleen and in lung macrophages. Its level is elevated in the brain tissue of those with Alzheimer's disease and Down syndrome. Cathepsin S may function in the processing of amyloid precursor protein to amyloid beta peptides.

Biological Activity
  • 183.8 mU/mg (before freezing), using method described by Kirsche &,

  • Z-Val-Arg-NHMec in an assay buffer of 0.1M KPO4, 5mM disodium EDTA,
  • pH 6.5, at 25AC, for 5 minutes, giving a change in fluorescence of 40, 000
  • fluorescence units. The assay is standardized so that a 0.1 M solution of
  • free 7-amino-4-methylcoumarin gives a reading of 1000 fluorescence units
