FAK1, a non-receptor protein-tyrosine kinase, orchestrates diverse cellular processes. It regulates cell migration, adhesion, spreading, cytoskeleton reorganization, focal adhesion dynamics, cell protrusion, cell cycle, proliferation, apoptosis, embryonic and placenta development, angiogenesis, cardiomyocyte migration, heart development, axon growth, neuronal migration, axon branching, synapse formation, osteogenesis, osteoblast differentiation, integrin signaling, and downstream signaling of growth factor receptors. FAK1 forms complexes with SRC and family members, activating multiple signaling pathways, influencing PI3K, AKT1, MAP kinase, GEFs, GAPs, Rho GTPases, RAC1, P53/TP53, ubiquitination, and proteasomal degradation. It phosphorylates substrates like NEDD9, SRC, ACTN1, ARHGEF7, GRB7, RET, WASL, PXN, STAT1, BCAR1, GIT2, and SHC1. Isoform 6 (FRNK) inhibits FAK1 phosphorylation and signaling, limiting LPXN nuclear accumulation and SRF-dependent gene transcription.
