Description: CAL-130 is a novel, potent and selective PI3Kdelta and PI3Kgamma inhibitor with potential anticancer activity and with IC50s of 1.3 and 6.1 nM, respectively. Constitutive phosphoinositide 3-kinase (PI3K)/Akt activation is common in T cell acute lymphoblastic leukemia (T-ALL). Although four distinct class I PI3K isoforms (alpha, beta, gamma, delta) could participate in T-ALL pathogenesis, none has been implicated in this process. In the absence of PTEN phosphatase tumor suppressor function, PI3Kgamma or PI3Kdelta alone can support leukemogenesis, whereas inactivation of both isoforms suppressed tumor formation.
