Description
This gene, chemokine (C-C motif) ligand 14, is one of several CC cytokine genes clustered on 17q11.2. The CC cytokines are secreted proteins characterized by two adjacent cysteines. The cytokine encoded by this gene induces changes in intracellular calcium concentration and enzyme release in monocytes. Multiple transcript variants encoding different isoforms have been found for this gene. Read-through transcripts are also expressed that include exons from the upstream cytokine gene, chemokine (C-C motif) ligand 15, and are represented as GeneID: 348249.

For the quantitative determination of human C-C motif chemokine 14(CCL14) concentrations in serum, plasma, tissue homogenates.

Specificity:
This assay has high sensitivity and excellent specificity for detection of human CCL14. No significant cross-reactivity or interference between human CCL14 and analogues was observed.

Test Principle:
This BioAssay™ employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CCL14 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CCL14 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CCL14 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCL14 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Storage and Stability:
Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
