This reagent is used to determine the urea content in human serum.

Urea is catalyzed by urease to generate ammonia and carbon dioxide. The produced ammonia becomes to glutamic acid under the action of glutamate dehydrogenase (GLDH) with presence of 2 - Ketoglutaric acid and NADH. At the same time, NADH is oxidized to NAD. NADH has absorption peak at 340 nm. The decline rate of NADH absorbance is in direct proportion to urea concentration in the sample.

This assay kit is suitable for automatic or semi - automatic biochemical analyzer with 340nm and 405nm wavelength.

The sample should be fasting serum. Anticoagulants c o ntaining ammonium salt cannot be used. Samples testing should be completed at the same day after collected. Otherwise, the samples should be cryopreserved and avoid repeated freeze - thaw cycles. Urea in the samples remains stable for 2 days at 2 - 8C.
This kit comes with a calibrator.
