Description
Recognizes an epitope within the N-Terminal (NT) region of both MAP1LC3A (Microtubule-associated proteins 1A/1B light chain 3A/LC3A) and MAP1LC3B (Microtubule-associated proteins 1A/1B light chain 3B/LC3B), ubiquitin-like proteins and members of the MAP1LC3 family, which are widely used as reliable markers for the monitoring of autophagy.

LC3-I is the cytosolic form of LC3, which is converted into the active, membrane-bound form LC3-II, during the autophagy process. Tracking the level of conversion of LC3-I to LC3-II provides an indicator of autophagic activity, and levels of LC3-II in particular, correlate with the extent of autophagosome formation, due to its association with the autophagosome membrane.

Applications:
Suitable for use in Immunofluorescence and Western Blot. Other applications not tested.

Recommended Dilution:
Western Blot: 1:1000-1:5000, detects a band of ~14-15kD corresponding to LC3-II, and a band of ~17kD corresponding to LC3-I, in HeLa cell lysates.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
