Description
In mammals, TLR identified as type I transmembrane signaling receptors with pattern recognition capabilities, have been implicated in the innate host defense to pathogens. TLR2 has been identified as a receptor that is central to the innate immune response to lipoproteins of Gram-negative bacteria, several whole Gram-positive bacteria, as well as a receptor for peptidoglycan and lipoteichoic acid and other bacterial cell membrane products. A functional interaction between TLR2 and TLR6 in the cellular response to various bacterial products has been discovered. TLR2 is regarded as an essential receptor for many eubacterial cell wall components, including lipoproteins and peptidoglycan.

Applications:
Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation, Western Blot, Immunofluorescence and Functional Studies. Other applications have not been tested.

Recommended Dilutions:
Flow Cytometry: 2ug/3x10e5 cells; untreated as well as 0.4 % formaldehyde fixed cells can be used; Do not permeabilize the cells or use 4 % faraformaldehyde as fixative.
Immunofluorescence: 5-10ug/ml; HMECs were fixed with 4% paraformaldehyde in PBS, permeabilized and blocked before staining.
Western Blot: 1ug/ml for non-reduced PBMCs
Immunohistochemistry (Frozen): Fix with acetone; block with 3% H2O2. Positive Control: human tonsils. (Useful, but T8050-09E is preferred.)
Immunohistochemistry (Paraffin): 1ug/ml for 16 hours at 4°C. HOPE-fixed. Positive Control: human lung type II alveolar epithelial cells.
Immunoprecipitation: Lysed monocytes were immunoprecipitated with antibody-conjugated Sepharose.
Functional Studies: 1ug/5x10e7 cells; blocks TNFa release (induced by bacteria) from PBMCs.
Optimal dilution determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
