Quality Tested by Leinco Western Blotting: To detect Mouse IL-21 this polyclonal antibody can be used at a concentration of 0.1 - 0.2 µg/ml. This polyclonal antibody should be used in conjunction with compatible second-step reagents such as PN:G505 and a chromogenic substrate such as PN:T343. The detection limit for Mouse IL-21 is 1 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.
ELISA: This Mouse IL-21 polyclonal antibody may be used as a capture antibody at a recomended concetration of 0.2-0.8 µg/mL. An appropriate detection antibody would be PN:I787 (Anti-Mouse IL-21 – Biotin). The correct standard would be PN: I-453 (Recombinant Mouse IL-21).

Storage Conditions
The lyophilized antigen affinity purified polyclonal antibody can be stored desiccated at -20°C to -70°C for twelve months from date of receipt. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles. No detectable loss of activity was observed after six months.
Product Description
Interleukin 21 (IL-21) is a T cell-derived pleiotropic cytokine that has potent regulatory effects on cells of the immune system, including natural killer (NK) cells and cytotoxic T cells that can destroy virally infected or cancerous cells.1, 2 This cytokine induces cell division/proliferation in its target cells. IL-21 elicits its effects on immune cells by interacting with a cell surface receptor known as the interleukin 21 receptor, IL-21R, which is expressed in bone marrow cells and various lymphocytes. IL-21 is closely related to IL-2 and IL-153 and is a key factor in the transition between innate and adaptive immune responses.4 The critical role of IL21 in promoting humoral immune responses makes it an important focus of potential therapeutic interventions in conditions characterised by overproduction of pathogenic autoantibodies.5

Other Applications Reported In Literature
Neutralization: This antibody is useful for neutralization of Mouse IL-21 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Mouse IL-21 (at 250 ng/ml) is 1 - 15 µg/ml.
Immunocytochemistry: This antibody will detect IL-21 in cells. The working dilution is 5 - 15 μg/mL.

Manufacturer - Specificity
Goat Anti-Mouse Interleukin 21 (IL-21) recognizes Mouse IL-21. This antigen affinity purified polyclonal antibody was purified using a proprietary chromatographic technique that includes covalently immobilizing the antigen proteins or peptides to agarose based beads. This purification method enhances specificity, reduces nonspecific binding of extraneous IgG and provides you with the most reliable reagent available for your early discovery research.
