Actin Antibody (for nitrocellulose membrane loading control studies). Chickens were immunized with purified actin protein. After repeated injections, immune eggs were collected from the hens, and the IgY fractions were purified from the yolks. These IgY fractions were then affinity-purified using an agarose matrix to which the whole protein was attached. The final preparation of this product involved mixing affinity purified anti-actin antibody (final concentration of 25 ug/ml) with IgY fractions of anti-actin antibody (final concentration of 10 mg/ml) in PBS with 50% glycerol. This preparation was then filter-sterilized.

Our anti-actin antibody (made against rabbit muscle actin) recognizes mouse, rat and human actin proteins in SDS-polyacrylamide gel transfers onto nitrocellulose membranes. This loading control assures that similar amounts of total proteins were added to each of the lanes in the gels. Note that this product does not work in transfers using PVDF membranes.

Chickens were immunized with purified actin from rabbit muscle emulsified in Freund’s adjuvants. After multiple injections, eggs were collected from the hens, IgY fractions were prepared from the yolks, and then affinity-purified antibodies were prepared using actin conjugated to an agarose matrix. The final product is a mixture of both affinity-purified antibodies (25 µg/mL) and purified IgY (10 mg/mL), mixed with glycerol 1:1 (v/v) (to prevent freezing at -20°C), augmented with sodium azide and then filter-sterilized.
