Gene Full Name
Gamma-aminobutyric acid receptor subunit gamma-2
Background
Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system. There are two major classes of GABA receptors: the GABA-A and the GABA-AB subtype of receptors. GABA-A-Rs are important therapeutic targets for a range of sedative, anxiolytic, and hypnotic agents and are implicated in several diseases including epilepsy, anxiety, depression, and substance abuse. The GABA-A-R is a multimeric subunit complex. To date six ?s, four ?s and four ?s, plus alternative splicing variants of some of these subunits, have been identified (Olsen and Tobin, 1990; Whiting et al., 1999; Ogris et al., 2004). Injection in oocytes or mammalian cell lines of cRNA coding for ?- and ?-subunits results in the expression of functional GABA-A-Rs sensitive to GABA. However, coexpression of a ?-subunit is required for benzodiazepine modulation. It has recently been suggested that PKC? regulates the sensitivity of GABA-A ?1?2?2 receptors to ethanol and benzodiazepines through phosphorylation of serine 327 in the large intracellular loop of ?2 (Qi et al., 2007)
Immunogen
Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser327 of the ?2 subunit of rat GABAA receptor, conjugated to keyhole limpet hemocyanin (KLH). Immunogen species is Rat.
Contents
10 mM HEPES (pH 7.5), 150 mM NaCl, 100 µg per ml BSA and 50% glycerol.
Purification
Prepared from pooled rabbit serum by affinity purification via sequential chromatography on phospho and non-phosphopeptide affinity columns.
Concentration
0.5-1mg/ml, actual concentration vary by lot. Use suggested dilution ratio to decide dilution procedure.
Phospho Site
Phosphorylated, Ser327

Protein Name
Gamma-aminobutyric acid receptor subunit gamma-2
Protein Function
Component of the heteropentameric receptor for GABA, the major inhibitory neurotransmitter in the vertebrate brain. Functions also as histamine receptor and mediates cellular responses to histamine. Functions as receptor for diazepines and various anesthetics, such as pentobarbital; these are bound at a separate allosteric effector binding site. Functions as ligand- gated chloride channel.
Subcellular Localization
Cell junction, synapse, postsynaptic cell membrane; Multi-pass membrane protein. Cell membrane; Multi-pass membrane protein.
