Storage Conditions
Store lyophilized protein at -20C. Aliquot the product after reconstitution to avoid repeated freezing/thawing cycles. Reconstituted protein can be stored at 2-4C for a limited period of time. The lyophilized protein remains stable until the expiration date when stored at -20C.
Description
Osteocrin is a recently identified secreted protein expression of which was only detected in bone, peaking just after birth and decreasing markedly with age. It is a 103 aa protein with a MW = 11.4 kDa. Research studies indicate that osteocrin represents a novel, unique vitamin D-regulated bone-specific protein that appears to act as a soluble osteoblast regulator. In primary osteoblastic cell cultures, osteocrin expression coincides with matrix formation then decreases in very mature cultures. Treatment of cultures with 1, 25-dihydroxyvitamin D3 results in a rapid dose- dependent down-regulation of osteocrin expression, suggesting direct regulation. Chronic treatment of primary cultures with osteocrin-conditioned media inhibits mineralization and reduces osteocalcin and alkaline phosphatase expression. Recombinant Human Osteocrin is produced with an N-terminal His Tag. The His-Tagged Fusion Protein is 13.6 kDa containing 106 aa residues of Osteocrin and a 16 aa His Tag.
Formulation
Filtered (0.4 um) and lyophilized from 0.5 mg/ml in 0.05M Acetate buffer, pH 4.0.
Reconstitution
It is recommended to add 0.1 M Acetate buffer, pH 4, to prepare a working stock solution of approximately 0.5 mg/ml and to let the lyophilized pellet dissolve completely. For conversion to a higher pH value, we recommend intensive dilution by the relevant buffer to a concentration of 10 ug/ml. At higher concentrations, the solubility of this antigen is limited. Product is not sterile! Please filter the product by an appropriate sterile filter before using it in cell culture.
