Description of target
Key component of innate and adaptive immunity. TLRs (Toll-like receptors) control host immune response against pathogens through recognition of molecular patterns specific to microorganisms. TLR9 is a nucleotide-sensing TLR which is activated by unmethylated cytidine-phosphate-guanosine (CpG) dinucleotides. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response (PubMed:11564765, PubMed:17932028). Controls lymphocyte response to Helicobacter infection (By similarity). Upon CpG stimulation, induces B-cell proliferation, activation, survival and antibody production (PubMed:23857366).

Kit principle
The microplate provided in this kit has been pre-coated with an antibody specific to Toll Like Receptor 9 (TLR9). Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to Toll Like Receptor 9 (TLR9). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is proportional to the Toll Like Receptor 9 (TLR9) level in the sample or standard.

Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Toll Like Receptor 9 (TLR9) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Toll Like Receptor 9 (TLR9) were tested on 3 different plates, 8 replicates in each plate.
