Description of target	Adapter protein (clathrin-associated sorting protein (CLASP)) required for efficient endocytosis of the LDL receptor (LDLR) in polarized cells such as hepatocytes and lymphocytes, but not in non-polarized cells (fibroblasts). May be required for LDL binding and internalization but not for receptor clustering in coated pits. May facilitate the endocytocis of LDLR and LDLR-LDL complexes from coated pits by stabilizing the interaction between the receptor and the structural components of the pits. May also be involved in the internalization of other LDLR family members. Binds to phosphoinositides, which regulate clathrin bud assembly at the cell surface. Required for trafficking of LRP2 to the endocytic recycling compartment which is necessary for LRP2 proteolysis, releasing a tail fragment which translocates to the nucleus and mediates transcriptional repression (By similarity).

Kit principle	The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Low Density Lipoprotein Receptor Adaptor Protein 1 (LDLRAP1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Low Density Lipoprotein Receptor Adaptor Protein 1 (LDLRAP1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Low Density Lipoprotein Receptor Adaptor Protein 1 (LDLRAP1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Low Density Lipoprotein Receptor Adaptor Protein 1 (LDLRAP1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Kit reproducibility	Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Low Density Lipoprotein Receptor Adaptor Protein 1 (LDLRAP1) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Low Density Lipoprotein Receptor Adaptor Protein 1 (LDLRAP1) were tested on 3 different plates, 8 replicates in each plate.
