Description of target
A triglyceride (TG, triacylglycerol, TAG, or triacylglyceride) is an ester derived from glycerol and three fatty acids. There are many triglycerides, depending on the oil source, some are highly unsaturated, some less so. The chain lengths of the fatty acids in naturally occurring triglycerides vary, but most contain 16, 18, or 20 carbon atoms. Triglycerides, as major components of very-low-density lipoprotein (VLDL) and chylomicrons, play an important role in metabolism as energy sources and transporters of dietary fat.

Kit principle
Aviva Systems Biology Triglyceride ELISA Kit () (OKEH02597) is based on a competitive enzyme immunoassay technique. The microtiter well-plate in this kit has been pre-coated with an anti-Triglyceride antibody. Sample or standards are added to the wells along with a fixed quantity of biotinylated Triglyceride and incubated. The Triglyceride found in the sample or standards competes with the biotinylated Triglyceride for limited binding sites on the immobilized anti-Triglyceride antibody. Excess unbound biotinylated Triglyceride and sample or standard Triglyceride is washed from the plate. Avidin-HRP conjugate is added, incubated and washed. An enzymatic reaction is then produced through the addition of TMB substrate which is catalyzed by the immobilized HRP to generate a blue color product that changes yellow after adding acidic stop solution. The density of yellow coloration is measured by reading the absorbance at 450 nm which is quantitatively proportional to the amount of biotinylated Triglyceride captured in the well and inversely proportional to the amount of Triglyceride which was contained in the sample or standard.
