Induces KLC1 association with vesicles and functions as a cargo in axonal anterograde transport. Complex formation with APBA2 and APP, stabilizes APP metabolism and enhances APBA2-mediated suppression of beta-APP40 secretion, due to the retardation of intracellular APP maturation. In complex with APBA2 and C99, a C-terminal APP fragment, abolishes C99 interaction with PSEN1 and thus APP C99 cleavage by gamma-secretase, most probably through stabilization of the direct interaction between APBA2 and APP. The intracellular fragment AlcICD suppresses APBB1-dependent transactivation stimulated by APP C-terminal intracellular fragment (AICD), most probably by competing with AICD for APBB1-binding. May modulate calcium-mediated postsynaptic signals.

Assay Methodology: Quantitative Sandwich ELISA

Aviva Systems Biology CLSTN1 ELISA Kit (Human) (OKEI00311) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody specific for CLSTN1 has been pre-coated onto a 96-wellplate (12 x 8 Well Strips). Standards or test samples are added to the wells, incubated and removed. A biotinylated detector antibody specific for CLSTN1 is added, incubated and followed by washing. Avidin-Peroxidase Conjugate is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of a luminol substrate which is catalyzed by the HRP to produce light emission. The light emission is read by a luminometer (or photo-multiplier equipped instrument) and the intensity of the emitted light is proportional to the amount of sample CLSTN1
