CENPA

Histone H3-like nucleosomal protein that is specifically found in centromeric nucleosomes. Replaces conventional H3 in the nucleosome core of centromeric chromatin at the inner plate of the kinetochore. The presence of CENPA subtly modifies the nucleosome structure and the way DNA is wrapped around the nucleosome and gives rise to protruding DNA ends that are less well-ordered and rigid compared to nucleosomes containing histone H3. May serve as an epigenetic mark that propagates centromere identity through replication and cell division.

Assay Methodology: Quantitative Sandwich ELISA

Aviva Systems Biology Cenpa ELISA Kit (Mouse) (OKEI00396) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody specific for Cenpa has been pre-coated onto a 96-wellplate (12 x 8 Well Strips). Standards or test samples are added to the wells, incubated and removed. A biotinylated detector antibody specific for Cenpa is added, incubated and followed by washing. Avidin-Peroxidase Conjugate is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of a luminol substrate which is catalyzed by the HRP to produce light emission.
