Description of target

Ki-67(Proliferation-related Ki-67 antigen), also known as MKI67 or KIA, is a protein that in humans is encoded by the MKI67 gene. This gene is mapped to 10q26.2. From study of a panel of human-rodent somatic cell hybrids, it has been demonstrated that a gene involved in the expression of the MKI67 antigen is located on chromosome 10. By in situ hybridization, Fonatsch et al. (1991) regionalized the MKI67 gene to chromosome 10q25-qter. By FISH, Traut et al. Antigen KI-67 is a nuclear protein that is associated with and may be necessaryfor cellular proliferation. Furthermore it is associated with ribosomal RNA transcription. Inactivation of antigen KI-67 leads to inhibition of ribosomal RNA synthesis.

Kit principle

Aviva's human Ki67 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Ki67 has been precoated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Ki67 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The absorbance of yellow is proportional to the human Fibronectin amount of sample captured in plate.
