Description of target
This magnesium-dependent enzyme catalyzes the hydrolysis of ATP coupled with the translocation of calcium from the cytosol to the sarcoplasmic reticulum lumen. Isoform 2 is involved in the regulation of the contraction/relaxation cycle (PubMed:16402920). Acts as a regulator of TNFSF11-mediated Ca(2+) signaling pathways via its interaction with TMEM64 which is critical for the TNFSF11-induced CREB1 activation and mitochondrial ROS generation necessary for proper osteoclast generation. Association between TMEM64 and SERCA2 in the ER leads to cytosolic Ca (2+) spiking for activation of NFATC1 and production of mitochondrial ROS, thereby triggering Ca (2+) signaling cascades that promote osteoclast differentiation and activation (By similarity).

Kit principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to ATPase, Ca++ Transporting, Cardiac Muscle, Slow Twitch 2 (ATP2A2). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to ATPase, Ca++ Transporting, Cardiac Muscle, Slow Twitch 2 (ATP2A2). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain ATPase, Ca++ Transporting, Cardiac Muscle, Slow Twitch 2 (ATP2A2), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of ATPase, Ca++ Transporting, Cardiac Muscle, Slow Twitch 2 (ATP2A2) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level ATPase, Ca++ Transporting, Cardiac Muscle, Slow Twitch 2 (ATP2A2) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level ATPase, Ca++ Transporting, Cardiac Muscle, Slow Twitch 2 (ATP2A2) were tested on 3 different plates, 8 replicates in each plate.
