The protocol enables detection of antigens in intraellular and extracellular locations.

Stock solutions can be thawed-frozen multiple times, or aliquoted and frozen.
Cells are first washed with the provided PBS solution, and then fixed with a paraformaldehyde-PBS solution (paraformaldehyde provided by user). The reaction is quenched with the provided quenching buffer. Subsequently cells are permeabilized with the provided permeabilization buffer and developed with 1 to 3 primary antibodies simultaneously in the provided detection buffer. The unbound primary antibodies are washed away in the provided detection buffer and then cells are incubated again with 1 to 3 secondary antibodies that are conjugated with distinct fluorescent tags. After washes with detection buffer, the microscope slides are preserved in anti-fade solution (Fivephoton Biochemicals Part No. AF-1).
The immunofluorescence labeling kit employs saponin to permeabilize membranes. Saponin more effectively preserves membrane structure, enhancing resolution of intracellular organelles in fluorescent microscopy images.

Along with intracellular antigens, transmembrane and cell surface proteins can also be visualized with immunofluorescence protocols that employ membrane permeabilization since antibodies are accessible to all cellular compartments and locations.
