Immunogen:
Human B7-H4
Specificity:
Recognises human B7-H4 also known as B7x a costimulatory protein which is reported to function as a negative regulator of T-cell mediated immunity. Although B7-H4 binds an unknown receptor it is thought to deliver an inhibitory signal to T-cells preventing their proliferation cell cycle progression and interleukin-2 production. B7-H4 deficient mice are only minimally affected; suggesting B7-H4 is important in the fine tuning of the T-cell mediated immune response. B7-H4 is expressed on activated T-cells B-cells monocytes and dendritic cells. Aberrant expression has been associated with cancers of the lung breast and ovary in humans. B7-H4 could be a useful prognostic marker in Renal Cell Carcinoma (RCC). Recommended Negative Controls: MOUSE IgG1 NEGATIVE CONTROLRecommended Secondary Antibodies: Rabbit Anti Mouse IgGGoat Anti Mouse IgGGoat Anti Mouse IgG (H/L)Goat Anti Mouse IgG IgA IgMHuCAL Anti Mouse IgG1Goat Anti Mouse IgG (Fc)Sheep Anti Mouse IgG (H/L)
Function:
Negatively regulates T-cell-mediated immune response by inhibiting T-cell activation proliferation cytokine production and development of cytotoxicity. When expressed on the cell surface of tumor macrophages plays an important role together with regulatory T-cells (Treg) in the suppression of tumor-associated antigen-specific T-cell immunity. Involved in promoting epithelial cell transformation. Ref. 12Ref. 13Ref. 16Ref. 17UniProtKB Q7TSP5Subcellular locationCell membrane; Single-pass type I membrane protein Potential.
Note:
Expressed at the cell surface. A soluble form has also been detected. Ref. 12Ref. 1Ref. 3Tissue specificityOverexpressed in breast ovarian endometrial renal cell (RCC) and non-small-cell lung cancers (NSCLC). Expressed on activated T- and B-cells monocytes and dendritic cells but not expressed in most normal tissues (at protein level). Widely expressed including in kidney liver lung ovary placenta spleen and testis. Ref. 12Ref. 13Ref. 17Ref. 1Ref. 3Ref. 11Ref. 15Induction: Up-regulated by IL-6 and IL-10 and inhibited by GM-CSF and IL-4 on antigen-presenting cells (APCs). Ref. 13Ref. 16Ref. 14Post-translational modificationN-glycosylated. Ref. 12Miscellaneous: May serve as a predictive marker for renal cell carcinoma. Ref. 15Sequence similaritiesBelongs to the immunoglobulin superfamily. BTN/MOG family. UniProtKB Q5ZPR3Contains 2 Ig-like V-type (immunoglobulin-like) domains. Caution: The mouse ortholog has been shown (PubMed:12818166) to be a GPI-anchored protein but the Gly residue which is predicted to be the modified site in mouse and rat is not conserved in human.
