Application Note:
WB: Use at a concentration of 2-4 µ g/ml. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. The caspase family of cysteine proteases has been shown to play a key role in apoptosis. Caspase 6 is one of the major executioner caspases functioning in cellular apoptotic processes. Similar as other caspases Caspase 6 is also synthesized as an inactive pro-enzyme that is processed in cells undergoing apoptosis. Together with Caspase 3 Caspase 6 is one of the major caspases in apoptotic cells and functions downstream of apoptosis inhibitors Bcl-2 and Bcl-xL. Caspase 6 has also been shown involving in the proteolysis of poly (ADP-ribose) polymerase (PARP) and nuclear lamin A.
Function:
Involved in the activation cascade of caspases responsible for apoptosis execution. Cleaves poly(ADP-ribose) polymerase in vitro as well as lamins. Overexpression promotes programmed cell death.
Catalytic Activity:
Strict requirement for Asp at position P1 and has a preferred cleavage sequence of Val-Glu-His-Asp-|-.
Enzyme Regulation:
Activation is suppressed by phosphorylation at Ser-257.
Subunit:
Heterotetramer that consists of two anti-parallel arranged heterodimers each one formed by a 18 kDa (p18) and a 11 kDa (p11) subunit.
Subcellular Location:
Cytoplasm.
Ptm:
Cleavages by caspase-3 caspase-8 or -10 generate the two active subunits.
