Immunogen:
A peptide corresponding to a 28 amino acid sequence from near the centre of Human BRSK1.
Specificity:
Recognises human BR serine-threonine kinase 1 (BRSK1) a cytoplasmic and nuclear protein belonging to the protein kinase super family. BRSK1 is a widely expressed protein with highest levels being detected in the brain and testis. It is essential for the polarization forebrain neurones which results in the distinct properties of axons and dendrites. Research suggests that BRSK1 is involved in the regulation of G2/M arrest and functions as a checkpoint kinase following UV- or methyl methane sulfonate (but not IR-) induced DNA damage. BRSK1 phosphorylates WEE1 and CDC25B in vitro and CDC25C in vitro and in vivo. Multiple isoforms of BRSK1 have been identified. Recommended Secondary Antibodies: Sheep Anti Rabbit IgG
Function:
Required for the polarization of forebrain neurons which endows axons and dendrites with distinct properties possibly by locally regulating phosphorylation of microtubule-associated proteins By similarity. May be involved in the regulation of G2/M arrest in response to UV- or methyl methane sulfonate (MMS)-induced but not IR-induced DNA damage. Phosphorylates WEE1 and CDC25B in vitro and CDC25C in vitro and in vivo. Ref. 1Ref. 3Catalytic activityATP + a protein = ADP + a phosphoprotein. Cofactor: Magnesium. Enzyme regulationActivated by phosphorylation on Thr-205 by STK11 in complex with STE20-related adapter-alpha (STRAD alpha) pseudo kinase and CAB39. Ref. 1Subcellular locationCytoplasm. Nucleus.
Note:
Nuclear in the absence of DNA damage. Translocated to the nucleus in response to UV- or MMS-induced DNA damage. Ref. 3Tissue specificityWidely expressed with highest levels in brain and testis. Protein levels remain constant throughout the cell cycle. Ref. 3Sequence similaritiesBelongs to the protein kinase superfamily. CAMK Ser/Thr protein kinase family. AMPK subfamily. Contains 1 protein kinase domain. Contains 1 UBA domain. Sequence cautionThe sequence AAH16681. 1 differs from that shown. Reason: Erroneous initiation. 1. Cobbold S. & Metcalfe S.
