Specificity:
CSK
Immunogen:
Peptide sequence EQLEHIKTHELHL corresponding to the C-terminal region of CSK (NP_004374. 1).
Specificity Note:
This product specifically recognises an epitope within the C-terminal (CT) region of CSK (c-src tyrosine kinase) a cytoplasmic tyrosine kinase ubiquitously expressed as two mRNA species which down-regulates the activity of the c-src oncoprotein.
Immunohistology:
This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. TRIS/EDTA buffer pH 9. 0 is recommended for this purpose. Histology: Human Tonsil
Antiserum Preparation:
Antisera to human CSK (CT) were raised by repeated immunisations of goats with highly purified antigen. Purified IgG was prepared by affinity chromatography.
Buffer Solution:
TRIS buffered saline pH7. 3
Preservative Stabilisers:
0. 02%Sodium Azide0. 5%Bovine Serum AlbuminSuggested
Dilution:
Western Blot: This product detects a band of approximately 50kDa in Jurkat cell lysates. Suggested

Function:
Specifically phosphorylates \' Tyr-504\' on LCK which acts as a negative regulatory site. Can also act on the LYN and FYN kinases.
Catalytic Activity:
ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate.
Subunit:
Interacts with PTPN8 (By similarity). Interacts with phosphorylated SIT1 PAG1 LIME1 and TGFB1I1.
Subcellular Location:
Cytoplasm (By similarity). Cell membrane (By similarity). Note=Mainly cytoplasmic also present in lipid rafts (By similarity).
Tissue Specificity:
Expressed in lung and macrophages.
Ptm:
Autophosphorylation of Tyr-304 occurs only at abnormally high CSK concentrations in vitro.
Similarity:
Belongs to the protein kinase superfamily. Tyr protein kinase family. CSK subfamily.
Similarity:
Contains 1 protein kinase domain.
Similarity:
Contains 1 SH2 domain.
Similarity:
Contains 1 SH3 domain.
