Specificity:
UROKINASE
Specificity:
UROKINASE
Immunogen:
Peptide with sequence C-HFLPWIRSHTKEEN from the C-terminal of the Urokinase sequence.
Specificity Note:
This product detects an epitope within the C-terminal of urokinase a potent 50kD serine protease which is also known as urokinase plasminogen activator (uPA). uPA cleaves and activates plasminogen to form plasmin and is involved in extracellular cellular matrix degradation and cell signalling. Recent studies have implicated uPA and its receptor in the pathogenesis of inflammation and immunity. A specific polymorphism in the gene encoding uPA may be associated with late-onset Alzheimer\' s disease. The levels of uPA uPAR and its soluble fragments have also been found to be increased in tissues plasma and other body fluids of cancer patients and to be markers of cancer development and metastasis.
Antiserum Preparation:
Antisera to Urokinase were raised by repeated immunisations of goat with highly purified antigen. Purified IgG prepared by affinity chromatography.
Buffer Solution:
TRIS buffered saline pH7. 3
Preservative Stabilisers:
0. 02%Sodium Azide (NaN3)0. 5%Bovine Serum Albumin Suggested
Dilution:
Western Blot: This product detects a band of approximately 48kDa in KEH 293 cell lysates. Suggested
Dilution:
ELISA - 1:8000Western Blotting - 0. 3 - 1. 0ug/ml Summary: This protein is a serine protease involved in degradation of the extracellular matrix and possibly tumor cell migration and proliferation. A specific polymorphism in this gene may be associated with late-onset Alzheimer disease and also with decreased affinity for fibrin-binding. The protein encoded by this gene converts plasminogen to plasmin by specific cleavage of an Arg-Val bond in plasminogen. This protein\' s proprotein is cleaved at a Lys-Ile bond by plasmin to form a two-chain derivative in which a single disulfide bond connects the amino-terminal A-chain to the catalytically active carboxy-terminal B-chain. This two-chain derivative is also called HMW-uPA (high molecular weight uPA). HMW-uPA can be further processed into LMW-uPA (low molecular weight uPA) by cleavage of chain A into a short chain A (A1) and an amino-terminal fragment. LMW-uPA is proteolytically active but does not bind to the uPA receptor.
