Localization:
Cytoplasmic Cyclooxygenases (COX) are 72-kDa intralumenal residents of the endoplasmic reticulum (ER) and nuclear envelope where they catalyze the rate-limiting steps in the conversion of arachidonate to the physiologically dynamic prostanoids (Ballif et al. 1996). Numerous investigations have shown that the isoform COX-1 is not a participant in the pathway of inflammation process (Marnett 2000) and colon carcinogenesis (Prescott & Fitzpatrick 2000). Although research on COX-2 inhibitors has focused mainly on inflammation and pain experimental and epidemiological data suggest that NSAID (Non steroidal ant-inflammatory drugs) should be used without any effect on COX-1 in the treatment or prevention of a broader range of diseases (Marnett & Kalgutkar 1999).
Function:
May play an important role in regulating or promoting cell proliferation in some normal and neoplastically transformed cells.
Catalytic Activity:
Arachidonate + AH2 + 2 O2 = prostaglandin H2 + A + H2O. Cofactor: Binds 1 heme B (iron-protoporphyrin IX) group per subunit (By similarity).
Pathway:
Lipid metabolism; prostaglandin biosynthesis.
Subunit:
Homodimer.
Subcellular Location:
Microsome membrane; Peripheral membrane protein. Endoplasmic reticulum membrane; Peripheral membrane protein.
Miscellaneous:
This enzyme acts both as a dioxygenase and as a peroxidase.
Miscellaneous:
This enzyme is the target of nonsteroidal anti-inflammatory drugs such as aspirin.
Similarity:
Belongs to the prostaglandin G/H synthase family.
Similarity:
Contains 1 EGF-like domain.
